      DMEM, high glucose, GlutaMAX™ Supplement Gibco™

      DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12.       Using DMEM
      DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate. DMEM with GlutaMAX™ supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. DMEM contains no proteins, lipids, or growth factors. Therefore, DMEM requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). DMEM uses a sodium bicarbonate buffer system (3.7 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

      cGMP manufacturing and quality system
      DMEM is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards. For supply chain continuity, we offer an identical DMEM product made in our Scotland facility (61965-059). This facility is also registered with the FDA as a medical device manufacturer and certified to the ISO 13485 standard.

      Expi293F™ Inducible Cells Gibco™

      The Expi293F Inducible cell line is derived from engineered Expi293F cells that stably express the tetracycline repressor (TetR) protein, thus enabling tetracycline-regulated protein expression. The cell line was established by the stable integration of the regulatory plasmid pcDNA6/TR, which encodes the TetR gene under the control of the human CMV promoter. It is a powerful tool for tetracycline-inducible high-yield expression of recombinant proteins.

      Features of Expi293 Inducible cells include:
      • Suspension, high-density culture in Expi293 Expression Medium
      • Tetracycline-inducible protein expression with extremely low basal expression levels
      • Equivalent protein yields to parental Expi293F cells—up to 1 g/L
      • Significantly higher protein yields than other HEK293 Tet-inducible cell line alternatives

      For inducible expression of your protein of interest, pcDNA5/TO mammalian expression vector (Cat. No. V103320) or an equivalent inducible vector is required for tetracycline-regulated gene expression.

      Frozen cells are supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in Expi293 Expression Medium and 10% DMSO. The cells should be thawed directly into Expi293 Expression Medium.

      B-27™ Supplement, minus insulin Gibco™

      A variation of the most cited neuronal cell culture supplement, B-27 Supplement Minus Insulin is a customized version of the original B-27 Supplement without insulin. B-27 Supplement Minus Insulin is a serum-free supplement designed to support growth of hippocampal and other CNS neurons in applications where the presence of insulin causes interference. B-27 Supplement Minus Insulin is provided as a 50X liquid and is intended for use with Neurobasal Medium or Neurobasal-A Medium.

      B-27 Supplement is suitable for use in most neuronal cell applications. For other applications, we provide alternative formulations of B-27 Supplement and Neurobasal media.

      Collagenase, Type IV, powder Gibco™

      Collagenase is a protease that cleaves the bond between a neutral amino acid (X) and glycine in the sequence Pro-X-Glyc-Pro, which is found with high frequency in collagen. Collagenase is unique among proteases in its ability to degrade the triple-helical native collagen fibrils commonly found in connective tissues such as skin, tendon, blood vessels, and bone. Collagenase disaggregation is suitable for the culture of human tumors, mouse kidney, human adult and fetal brain, and many other tissues including epithelium. Collagenase is relatively gentle, dissociates well at physiological temperature and pH, and requires no mechanical agitation or special equipment.

      Gibco® Collagenase Type IV is isolated from Clostridium histolyticum and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation and organ perfusions. Gibco® Collagenase Type IV activity is guaranteed to be greater than 160 units/mg. Compared to other collagenase preparations, Gibco® Collagenase Type IV has low tryptic activity and is well-suited for the digestion of islet cells from the pancreas.

      Dynamis™ AGT™ Medium Gibco™

      Gibco® Dynamis™ AGT™ dry format medium is specifically designed to offer the highest batch and fed-batch culture performance and yield with recombinant CHO cells in a chemically defined environment.       Expi293F™ Inducible GnTI- Cells Gibco™

      The Expi293F Inducible GnTI- cell line is derived from engineered Expi293F cells that do not have N-acetylglucosaminyltransferase I (GnTI) activity, therefore lacking complex N-glycans, and stably express the tetracycline repressor (TetR) protein, thus enabling tetracycline-regulated protein expression. The cell line was established by the selective knockout of the GnTI gene and stable integration of the regulatory plasmid pcDNA6/TR, which encodes the TetR gene under the control of the human CMV promoter. It is a powerful tool for tetracycline-inducible high-yield expression of homogeneously glycosylated recombinant proteins.

      Features of Expi293F Inducible GnTI- cells include:

      • Homogeneous N-glycosylation of expressed proteins

      Gibco® FluoroBrite™ DMEM features a background fluorescence that is comparable to PBS and 90% lower than that emitted by standard phenol red–free DMEM. Formulated to include the required nutrients for routine cell culture when supplemented with 10% fetal bovine serum and 4 mM L-glutamine or GlutaMAX™ Supplement, FluoroBrite™ DMEM is designed to enhance the signal-to-noise ratio of fluorophores, enabling researchers to visualize even the weakest fluorescent events in an environment that promotes optimum cell health. Additional features include:

      Live-cell fluorescence microscopy is an essential technique for the visualization of fundamentally important and physiologically relevant biological events. A key challenge with this technique is the ability to image weak fluorophores without causing cell damage, photobleaching, or undesirable changes to cell health. FluoroBrite™ DMEM helps address these issues.

      Dispase II, powder Gibco™

      Dispase II (neutral protease) is an amino-endo peptidase that hydrolyzes the N-terminal peptide bonds of non-polar amino acid residues. This sequence is found with high frequency in collagen. Dispase II has a mild proteolytic action that makes it useful for the isolation and routine passaging of primary cells. Dispase II is relatively gentle, dissociates well at physiological temperature and pH, and generally maintains cell membrane integrity.

      Gibco® Dispase II is produced in Bacillus polymyxa and packaged as a lyophilized, non-sterile powder for research use in cell or tissue dissociation, and to prevent clumping in suspension cultures. Gibco® Dispase II activity is guaranteed to be greater than 0.5 units/mg.

      MCDB 131 Medium, no glutamine Gibco™

      MCDB 131 Medium was originally developed by Knedler and Ham as reduced serum-supplemented medium for the culture of human microvascular endothelial cells (HMVEC). MCDB 131 Medium is also used with other cell types, including human omental microvascular cells, hepatocytes, myocytes, and smooth muscle cells.

      Using MCDB 131
      MCDB 131 Medium contains many components not found in traditional basal media, such as trace elements, putrescine, adenine, thymidine, and higher levels of some amino acids and vitamins. These additions allow the medium to be supplemented with very low levels of serum or defined components. MCDB 131 Medium contains no proteins or growth factors, and is often supplemented with EGF, hydrocortisone, glutamine, and low levels of Fetal Bovine Serum (FBS). The FBS concentration must be optimized for each cell type. MCDB 131 Medium uses a sodium bicarbonate buffer system (1.176 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

      Dual-site cGMP manufacturing
      For supply chain continuity, we manufacture MCDB 131 at two separate facilities, located in Grand Island, NY and Scotland, UK. Both sites are compliant with cGMP-manufacturing requirements, are certified to ISO 13485, and are registered with the FDA as medical device manufacturers.

      Protein Expression Medium (PEM) Gibco™

      Ham's F-10 Nutrient Mix Gibco™

      Ham's F-10 Nutrient Mixture (F-10) was designed for serum-free growth of Chinese Hamster Ovary (CHO) cells. F-10 has since been used for serum-free growth of CHO cultures as well as serum-supplemented growth of other mammalian cells, including COS-7, primary rat astrocytes, and rat prostate epithelial cells.

      Using Ham's F-10
      Compared to other basal media, F-10 contains a wider variety of components, including zinc, hypoxanthine, and thymidine. Serum-free growth of CHO cells in F-10 has led to a variety of improved formulations, such as Ham's F-12 Nutrient Mixture (F-12). F-10 contains no proteins or growth factors. Therefore, F-10 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). F-10 uses a sodium bicarbonate buffer system (1.2 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.
