                The Molecular Probes® BioParticles® product line consists of a series of fluorescently labeled, heat- or chemically killed bacteria and yeast in a variety of sizes, shapes, and natural antigenicities. These fluorescent BioParticles® products have been employed to study phagocytosis by fluorescence microscopy, quantitative spectrofluorometry, and flow cytometry.

                We offer E. coli (K-12 strain), S. aureus (Wood strain, without protein A) and zymosan (S. cerevisiae) BioParticles® conjugates covalently labeled with a variety of different fluorophores (special care has been taken to remove free dye after conjugation). Unlike the fluorescence of fluorescein-labeled BioParticles® conjugates, which is partially quenched in acidic environments, the fluorescence of the Alexa Fluor®, BODIPY® FL, tetramethylrhodamine and Texas Red® dye conjugates is uniformly intense over the pH range from 3 to 10.

                Using BioParticles Products
                BioParticles® conjugates are provided as lyophilized powders. There are approximately 3 x 108 E. coli or S. aureus particles per mg solid and approximately 2 x 107 zymosan particles per mg solid. BioParticles® conjugates can be reconstituted in the buffer of your choice for use in phagocytosis assays. The fluorescence of BioParticles® conjugates that are bound to the surface of the cell (but not internalized) can be quenched by ethidium bromide, trypan blue, or other quenchers. In addition to cellular applications, fluorescent BioParticles® conjugates may be effective as flow cytometry calibration references when sorting bacteria and yeast mutants. These small particles may also be useful references for light scattering studies because their sizes and shapes differ in characteristic ways.

This supplement is a mixture of peptides, amino acids, and carbohydrates, both simple and complex. Customers have successfully used Bacto TC Lactalbumin Hydrolysate to support the growth of many mammalian, bacterial, and insect cell types, and it has been used to produce both recombinant proteins and animal health vaccines. Customers have also used this supplement as an amino acid supplement for mammalian cell culture media.

                EfficientFeed™ C+ 2X Supplement Gibco™

                Gibco EfficientFeed C+ 2X Supplement is a single-part liquid supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. This product is designed to work with CHO cell lines expressing endogenous glutamine synthetase (GS) as a single feed without the need for additional GS supplements. Compared to the original CD EfficientFeed C AGT nutrient supplement, EfficientFeed C+ 2X Supplement has components that are super-concentrated from 81.2 g/L (1X AGT) to 162.4 g/L (2X), helping reduce product dilution or increase titers with additional feeding. The product is an animal origin-free, chemically defined formulation that contains no proteins, hydrolysates, or components of incompletely defined composition.

                Propidium Iodide Ready Flow™ Reagent Invitrogen™

                Propidium Iodide Ready Flow™ Reagent is a bright, easy-to-use nucleic acid stain for distinguishing dead cells from live cells in flow cytometry assays. This dye has excitation/emission maxima of 535/617 nm.

                Propidium Iodide Ready Flow Reagent is a commonly used dead-cell stain that emits red fluorescence when bound to DNA. Add 2 drops from the dropper bottle directly into 1 x 106 cells, incubate, and analyze.

                Propidium iodide is membrane impermeant and therefore excluded from viable cells. It binds to DNA by intercalating between the bases, thereby binding both DNA and RNA. Propidium iodide is commonly excited with the 488 nm laser, but others such as the 532 and 561 nm lasers are also suited for PI excitation.

                Poly-D-Lysine Gibco™

                Poly-D-Lysine is a chemically synthesized extracellular matrix used to facilitate cell adhesion to tissue culture-treated plastic and glass surfaces. Poly-D-Lysine contains polymers with a molecular weight of 50,000–150,000 Daltons, which makes this product ideal for neuronal culture applications. This ready-to-use product is provided as a liquid at a concentration of 0.1 mg/mL. When used on Nunc cell culture plastics with B-27 supplements or the B-27 Plus Neuronal Culture System, Poly-D-Lysine maintains cell viability and normal growth characteristics in multiple neuronal cell types.

                Epidermal Growth Factor, Biotinylated, complexed to Alexa Fluor™ 488 Streptavidin (Alexa Fluor™ 488 EGF complex) Invitrogen™

                We offer several conjugates of epidermal growth factor that are useful for the detection of EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. The conjugates incorporating fluorescein (FITC), Oregon Green® 514, and tetramethylrhodamine were constructed with the dye directly attached to EGF. The conjugates of EGF with Alexa Fluor® dyes and Texas Red® dye are complexes of the dye–streptavidin molecule and biotinylated EGF. The biotinylated conjugates utilize biotin-XX, which contains a long spacer arm designed to enhance the probe’s affinity for the EGF receptor.

                Geneticin™ Selective Antibiotic (G418 Sulfate), Powder Gibco™

                Geneticin® Selective Antibiotic (G418 Sulfate) is produced by the bacterium Micromonospora rhodorangea and acts by binding the ribosome, thus inhibiting protein synthesis in both prokaryotic and eukaryotic cells. Resistance to Geneticin® Selective Antibiotic (G418 Sulfate) is conferred by the E. coli APH (3’) – I and APH (3’) – II resistance genes.                 Gibco™ 2-Mercaptoethanol (also known as beta-mercaptoethanol or BME) is a potent reducing agent used in cell culture media to prevent toxic levels of oxygen radicals. 2-Mercaptoethanol is not stable in solution, so most protocols require daily supplementation. Gibco 2-Mercaptoethanol contains 2-mercaptoethanol at a concentration of 55 mM in Dulbecco's phosphate buffered saline (DPBS).
